I think if you want to set up a different lead sound, pumping more gain into the amp is a very poor idea. The MXR 10 band EQ is definetly the best way to go, especially seeing as you're happy with your rythm sound already.

I find a good lead sound has less gain than a good rythm sound. The MXR has additional volume and gain controls, so if you throw that in the loop, you could boost the signal, really shape your tone, and roll off some gain too, giving you a much more effective lead.

I won't disagree with Zimbloth about the tubescreamer tightening things up, but if you're talking about setting up a seperate lead sound, then it's not what you're looking for.
